Facts about Larotrectinib

Mechanism of Action

Larotrectinib is a strong inhibitor of TRK (TRK a, TRK b, TRK c). There are specific genes coding for each one– NTRK 1, NTRK 2, NTRK 3. Overactivity of the kinases usually results in the rapid growth of tumor cells, which could result from overexpression of any of the protein kinases in a cell. The fusion of the genes coding for these kinases may activate any of the kinases. Administration of larotrectinib, both in vivo and in vitro, has shown activities against tumor cell growth. Further studies have proven that the drug inhibits the protein kinases to cause diminished tumor cell growth.[2]
